Catalog Antibody> | The midgut samples from the control and phoxim-exposed groups were homogenized in lysis buffer supplemented with 1 mM of PMSF. Mitochondrial and cytosolic proteins were extracted with Mitochondrial Isolation Kit (Sangon, China) according to the manufacturer’s protocol. The samples were centrifuged at 15 800gfor 10 min, and the supernatants were collected. The following procedure was carried out following Gu et al. (2011). Antibody against α-Tubulin (Cell signaling, USA; 1:2000) and cytochrome c (Genscript, China; 1:2000) was used as the primary antibody, and the HRP-conjugated goat anti-rabbit IgG (1:10 000) was used as the secondary antibody. | Get A Quote |
The silkworm is an important economic insect. Poisoning of silkworms by organophosphate pesticides causes tremendous loss to the sericulture. In this study, Solexa sequencing technology was performed to profile the gene expression changes in the midgut of silkworms in response to 24 h of phoxim exposure and the impact on detoxification, apoptosis and immune defense were addressed. The results showed that 254 genes displayed at least 2.0-fold changes in expression levels, with 148 genes up-regulated and 106 genes down-regulated. Cytochrome P450 played an important role in detoxification. Histopathology examination and transmission electron microscope revealed swollen mitochondria and disappearance of the crista... More
