The specificity of the MonoRab™ Anti-FMC63 Antibody (24B2), mAb, Rabbit was evaluated using Raji cells as the experimental system. In the first group, Raji cells were incubated with a 1:50 dilution of MonoRab™ Anti-FMC63 Antibody (24B2), mAb, Rabbit (GenScript, A02327) followed by staining with a PE conjugated anti-rabbit IgG antibody, showing a negative signal (blue line). In the second group, Raji cells were incubated with FMC63 scFv, followed by staining with the same concentration of MonoRab™ Anti-FMC63 Antibody (24B2), mAb, Rabbit (GenScript, A02327) and PE conjugated anti-rabbit IgG antibody, resulting in a positive signal (red line) .
MonoRab™ Anti-FMC63 Antibody (24B2), mAb, Rabbit binds to FMC63 scFv in ELISA assays.
Recombinant FMC63 scFv was coated at a concentration of 1 μg/mL, followed by the addition of MonoRab™ Anti-FMC63 Antibody (24B2), mAb, Rabbit (GenScript, A02327) at serially diluted concentrations starting from 1 μg/mL. Detection was performed using Mouse Anti-Rabbit IgG Fc Antibody (14H9H10)[HRP], mAb (GenScript, A01856) diluted at 1:20,000.
The detection of PBMCs with or without FMC63 CAR-T cells using iFluor 647 conjugated MonoRab™ Anti-FMC63 Antibody (24B2), mAb, Rabbit (GenScript, A02327) demonstrated minimal nonspecific binding to PBMCs and specific recognition of FMC63-expressing CARs.

The FMC63 scFv (single-chain variable fragment) is a key component in many chimeric antigen receptor (CAR) designs, particularly in CAR-T cell therapies. Derived from a murine monoclonal antibody, FMC63 specifically targets human CD19, a cell surface protein highly expressed on B-cells, including malignant B-cells in B-cell leukemias and lymphomas. This scFv fragment is engineered by fusing the variable regions of the heavy and light chains of the antibody, preserving the antigen-binding capability while significantly reducing its size. This smaller, single-chain format enhances its integration into CAR constructs, enabling the recognition and destruction of CD19-positive cells by CAR-T cells. FMC63-based CARs are widely used in immunotherapy, particularly in FDA-approved treatments for B-cell malignancies, such as Kymriah® and Yescarta®. |
